Products Description

What is 7-Dehydrocholesterol (7-DHC)?

7-Dehydrocholesterol (7-DHC) is a naturally occurring sterol compound with the chemical formula C₂₇H₄₄O and a molecular weight of 384.64 g/mol. Structurally, it is a derivative of cholesterol, distinguished by the presence of an additional double bond in the B-ring of the steroid nucleus. Specifically, 7-DHC contains conjugated double bonds at the C5–C6 and C7–C8 positions (a Δ⁵,⁷ conjugated diene system), whereas cholesterol has only a single double bond at the C5 position.

Its two most critical biological identities are:

The direct biological precursor to vitamin D₃ (cholecalciferol); and A key intermediate in the cholesterol biosynthetic pathway.

Biosynthesis and Metabolic Origin in the Human Body OF 7-Dehydrocholesterol (7-DHC)

1 Endogenous Synthesis

The human body synthesizes 7-DHC endogenously and does not rely entirely on dietary intake. The primary sites of synthesis are:

Keratinocytes in the epidermal basal layer of the skin; and Hepatocytes in the liver.

The biosynthetic pathway begins with acetyl-CoA, proceeding through the mevalonate pathway → squalene → lanosterol → a series of enzymatic transformations, ultimately yielding 7-DHC.

2 Role as a Cholesterol Biosynthetic Intermediate

7-DHC is the penultimate intermediate in the Bloch pathway of cholesterol synthesis. It is converted to cholesterol by the enzyme 7-dehydrocholesterol reductase (DHCR7) , which catalyzes the stereospecific reduction of the Δ⁷ double bond. This pathway operates in virtually all tissues throughout the body.

3 Conversion to Vitamin D₃ – The Most Important Biological Function

This is the most well-known and physiologically significant function of 7-DHC. When 7-DHC in the skin is exposed to UVB radiation (wavelength 290–320 nm) , the conjugated diene system undergoes a photochemical electrocyclic ring-opening reaction, cleaving the B-ring to form previtamin D₃. Over several hours to days at body temperature, previtamin D₃ undergoes a thermal isomerization (a [1,7]-sigmatropic hydrogen shift) to produce vitamin D₃ (cholecalciferol) .

Applications Of 7-Dehydrocholesterol (7-DHC)

1. Industrial synthesis precursor of vitamin D₃: About 100 tons of crystalline vitamin D₃ are produced globally every year, almost all of which use 7-DHC as the key intermediate. Industrial production usually starts from cholesterol in lanolin, synthesizes 7-DHC through multi-step chemical reactions, and then converts it into vitamin D₃ through ultraviolet irradiation.

2. The human body's own "vitamin D factory": In human skin, 7-DHC is the direct raw material for the synthesis of vitamin D₃. After receiving UVB radiation in the sun, 7-DHC in the epidermal layer of the skin converts into vitamin D₃ through a photochemical reaction. This is the main way for the human body to obtain vitamin D.

3. Skin care products and cosmetic raw materials: Studies have shown that 7-DHC can inhibit epidermal keratin formation and melanoma cell proliferation, so it is increasingly being used in the skin care and cosmetics industries.
4. Synthetic biology and new materials: In view of the importance of 7-DHC, the use of microorganisms (such as Saccharomyces cerevisiae) for efficient and low-cost biosynthesis is a research hotspot. Through genetic engineering, the fermentation yield of 7-DHC has achieved a huge breakthrough, reaching 4.28 g/L in a 10-liter fermenter, which is the highest level reported so far.

Benefits Of 7-Dehydrocholesterol (7-DHC)

1. Value for skin care and anti-aging
Clinical evidence: A phase III clinical trial of xeroderma and pruritus in older women (n=18, 83.3% aged 60-74 years) showed that after 4 weeks of daily topical application of 7-DHC-vitamin D₃ complex cream:
Skin hydration is significantly improved (p value is statistically significant)
Improved transepidermal water loss (TEWL) – enhanced skin barrier function
Elevated serum vitamin D levels - transdermal absorption into systemic circulation
High patient comfort and satisfaction, well tolerated
2. Protective effect against tissue ischemia-reperfusion injury (IRI)
7-DHC is embedded in the cell membrane through a conjugated diene structure, directly capturing free radicals, inhibiting phospholipid auto-oxidation, and protecting the plasma membrane and mitochondrial membrane from oxidative damage. Inhibiting the DHCR7 enzyme (increasing 7-DHC levels) can effectively slow down the progression of renal IRI and provide new intervention strategies for ferroptosis-related diseases (ischemic injury, neurodegenerative diseases).

FAQ

Q: What are the recommended storage conditions and shelf life Of 7-Dehydrocholesterol (7-DHC)?

A: 7-DHC must be stored at −20°C, protected from light, and preferably under inert gas (nitrogen or argon) to prevent oxidative degradation-10-11. The compound is highly susceptible to autoxidation due to its conjugated diene structure-its peroxidation rate is approximately 200 times faster than cholesterol. Stock solutions should be used within 6 months when stored at −80°C, or within 1 month at −20°C

Q: What solvents can dissolve 7-DHC?

A: 7-DHC is soluble in ethanol (e.g., 12.5–16.67 mg/mL) and chloroform, but is insoluble or only slightly soluble in DMSO (<1 mg/mL)-9-11-4. For in vivo formulations, protocols recommend sequential addition of solvents: 10% EtOH + 40% PEG300 + 5% Tween-80 + 45% saline, or 10% EtOH + 90% corn oil for animal studies

Q: What is the relationship between 7-DHC and Smith-Lemli-Opitz Syndrome (SLOS)??

A: SLOS is a genetic disorder caused by DHCR7 gene mutations, where the enzyme that converts 7-DHC to cholesterol is defective-6. This leads to 7-DHC accumulation in plasma and tissues while cholesterol synthesis is impaired-4. 7-DHC is the diagnostic biomarker for SLOS-elevated plasma levels confirm the condition. Research-grade 7-DHC is used in cell culture studies of patient fibroblasts to study cholesterol metabolism.

Q: How should I handle the vial upon receipt?

A: Briefly centrifuge (small vials: ~500 rpm for 3 min) before opening to recover material from walls/cap. Allow cold-shipped vials to reach room temperature in a desiccator to avoid moisture condensation.

Q: What purity grades are available for 7-DHC?

A:Standard grade is ≥95% (HPLC), with premium research grades up to ≥98–99%. CAS 434-16-2, MW 384.64 g/mol.
